CDK4/6 inhibitors are a class of targeted therapy drugs that block cyclin-dependent kinases 4 and 6, proteins that regulate cell cycle progression from G1 to S phase. In hormone receptor-positive breast cancer, these inhibitors prevent cancer cells from dividing and multiplying. Common CDK4/6 inhibitors include palbociclib, ribociclib, and abemaciclib, typically used in combination with hormone therapy for advanced or metastatic hormone receptor-positive, HER2-negative breast cancer. These drugs have significantly improved progression-free survival in appropriate patient populations.
