A CRISPR-based gene-activation approach that uses dCas9 fused to a transcriptional activator to upregulate gene expression without modifying the DNA sequence.

Full Definition

CRISPR activation (CRISPRa) uses dCas9 coupled to transcriptional activator domains—such as VP64, VPR, or the SAM system—to enhance transcription of a target gene. It is used to study gene function, model gain-of-function phenotypes, or therapeutically upregulate underexpressed genes. Editors should write it as 'CRISPRa' with a lowercase 'a'. It is the functional counterpart of CRISPRi; the two are often discussed together. Neither CRISPRa nor CRISPRi alter the underlying DNA sequence.
