A hormone response element (HRE) is a specific DNA sequence found in the regulatory regions of genes that serves as a binding site for hormone-receptor complexes, thereby mediating hormone-induced changes in gene expression. Understanding HREs is important in hormone testing contexts, particularly when interpreting the clinical significance of hormone measurements and their downstream effects. Different classes of hormones have distinct response elements: steroid hormones bind to nuclear receptors that recognize palindromic HRE sequences, while thyroid hormones interact with thyroid hormone response elements (TREs). Knowledge of HRE function helps clinicians understand why certain hormone levels may not correlate directly with clinical symptoms, as receptor sensitivity and HRE polymorphisms can affect tissue responsiveness to circulating hormones.
