Matrix effects occur when components of the biological sample (serum, plasma, or urine) interfere with the analytical measurement of hormones, leading to artificially elevated or suppressed results. Common matrix effects include protein binding alterations, viscosity changes, ionic strength variations, and the presence of interfering substances like heterophile antibodies, cross-reactive medications, or endogenous compounds. These effects are particularly problematic in immunoassays where binding proteins or similar molecules may compete with or enhance antibody-antigen interactions. Understanding matrix effects is crucial for proper result interpretation, especially when results are discordant with clinical presentation or when comparing results between different analytical platforms.
