Radioimmunoprecipitation assay (RIPA) was once the gold standard for hormone testing, utilizing radioactive isotopes as tracers to detect and quantify hormones through competitive binding assays. The method involves mixing patient samples with known amounts of radioactively labeled hormones and specific antibodies, then measuring the radioactivity to determine hormone concentrations. While RIPA provided excellent sensitivity and specificity, it has been largely phased out in favor of non-radioactive methods due to safety concerns, disposal requirements, and regulatory restrictions. Understanding RIPA remains important for interpreting historical data and validation studies.
