The formation of fibrous connective tissue around a tumor, creating a dense stromal response.

Full Definition

A desmoplastic reaction refers to the proliferation of fibrous connective tissue and collagen deposition that occurs in response to malignant infiltration. This stromal response creates a dense, firm texture around the tumor and can be palpated clinically or visualized on imaging studies. Desmoplastic reactions are particularly characteristic of certain tumor types, including pancreatic adenocarcinoma, invasive breast carcinoma, and some sarcomas. The fibrotic response can make surgical resection more challenging and may affect drug penetration into the tumor. Pathologically, this reaction helps distinguish invasive carcinoma from in situ disease.
