Kisspeptin is a neuropeptide encoded by the KISS1 gene, initially discovered for its role in suppressing tumor metastasis but later recognized as a critical regulator of reproductive function. Kisspeptin neurons, primarily located in the arcuate nucleus and anteroventral periventricular nucleus of the hypothalamus, provide the major stimulatory input to GnRH neurons through the kisspeptin receptor (GPR54/KISS1R). This peptide is essential for pubertal development, as mutations in either kisspeptin or its receptor cause hypogonadotropic hypogonadism. Kisspeptin levels fluctuate across the menstrual cycle and are influenced by metabolic status, stress, and photoperiod, making it a key integrator of internal and external cues affecting reproduction.
