Full Definition

Leukocyte trafficking refers to the highly regulated process by which white blood cells migrate from the systemic circulation into central nervous system tissues. This multi-step process involves selectin-mediated rolling, chemokine-mediated activation, integrin-mediated firm adhesion, and transmigration across the blood-brain barrier or blood-CSF barrier. Under physiological conditions, leukocyte trafficking into the CNS is minimal due to the barrier properties and immunoprivileged status. However, during neuroinflammation, this process becomes dysregulated, allowing pathogenic immune cells to enter and contribute to tissue damage. Understanding leukocyte trafficking mechanisms has led to important therapeutic targets.
