Lomustine is often used as a single agent or in combination regimens for recurrent glioblastoma, anaplastic gliomas, and certain pediatric brain tumors. The drug causes DNA cross-linking and has a delayed myelosuppressive effect, requiring careful monitoring of blood counts. Treatment cycles are typically spaced 6-8 weeks apart due to its prolonged bone marrow suppression.
