A synthetic control arm is a computationally derived comparison group created using historical patient data and statistical modeling techniques. In oncology clinical trials, synthetic control arms can serve as an alternative to traditional placebo-controlled studies, particularly in rare cancers where recruiting sufficient control patients is challenging. These virtual control groups are constructed by identifying and matching historical patients with similar characteristics to the treatment group. Advanced matching algorithms consider factors such as tumor characteristics, staging, prior treatments, and patient demographics. Regulatory agencies are increasingly accepting synthetic control arms as valid comparators for accelerated approval pathways.
