In cancer genomics, variant annotation systems integrate information from sources such as ClinVar, COSMIC, OncoKB, and literature databases to provide clinicians with the context needed to determine whether identified mutations are likely to be pathogenic, benign, or of uncertain significance. Modern annotation pipelines automatically process thousands of variants and flag those with potential clinical relevance for manual review by molecular pathologists or genetic counselors.
