Microtomy is the laboratory procedure in which a microtome—a precision cutting instrument—is used to slice embedded tissue blocks into sections typically 3–5 micrometres thick for routine histology, or thinner for electron microscopy. The quality of sections depends on blade sharpness, section thickness setting, and the consistency of the embedding medium. Artefacts introduced during microtomy, such as chatter, folds, or compression, can compromise diagnostic accuracy and must be noted in quality-control documentation. Editors should distinguish 'microtomy' (the technique) from 'sectioning' (a broader, less specific term) and from 'cryosectioning,' which applies to frozen tissue.
Usage
Usage note: Prefer 'microtomy' over the generic 'sectioning' in formal laboratory documents. Specify section thickness in micrometres (µm), not millimetres.
