Glucose toxicity describes the detrimental effects of sustained hyperglycemia on both pancreatic beta-cell function and insulin sensitivity in peripheral tissues, creating a vicious cycle that accelerates the progression of type 2 diabetes. Chronic exposure to elevated glucose levels impairs insulin secretion through mechanisms including oxidative stress, endoplasmic reticulum stress, and glucolipotoxicity, while simultaneously worsening insulin resistance in muscle, liver, and adipose tissue. This concept underlies the rationale for early aggressive glucose control to preserve beta-cell function and prevent irreversible metabolic deterioration.
