The Liraglutide Effect and Action in Diabetes: Evaluation of Cardiovascular Outcome Results (LEADER) trial was a landmark randomized controlled trial that evaluated cardiovascular outcomes with liraglutide versus placebo in patients with type 2 diabetes at high cardiovascular risk. The trial demonstrated a significant 13% reduction in major adverse cardiovascular events with liraglutide treatment. LEADER was one of the first cardiovascular outcomes trials to show cardiovascular benefits with a GLP-1 receptor agonist, leading to regulatory approval for cardiovascular indication. The trial's positive results fundamentally changed diabetes treatment paradigms and influenced subsequent guideline recommendations for patients with established cardiovascular disease or high cardiovascular risk.
