Tandem mass spectrometry (MS/MS) employs two or more mass analyzers in sequence to provide exceptional specificity and sensitivity for hormone measurement. The first analyzer selects the target molecule, which is then fragmented, and the second analyzer identifies characteristic fragments. This technique eliminates cross-reactivity issues common in immunoassays and is considered the gold standard for measuring steroid hormones, particularly when concentrations are low or when structural analogs may interfere with immunoassay methods.
