Full Definition

A specific type of abnormal neuron characterized by marked cytoplasmic swelling, loss of normal Nissl substance, and a pale, vacuolated appearance that gives the cell a balloon-like morphology. These neurons are most characteristically associated with corticobasal degeneration but can also be observed in other tau-positive neurodegenerative diseases including Alzheimer's disease and Pick's disease. Balloon neurons typically show immunoreactivity for phosphorylated neurofilaments and may contain tau inclusions, representing a form of neuronal stress response to protein aggregation and cellular dysfunction. They are most commonly found in the frontal and parietal cortex, particularly in superficial cortical layers. In neuropathological diagnosis, the identification of balloon neurons can provide important clues about the underlying disease process and helps distinguish between different tauopathies.
