Somatotrophs are acidophilic cells located in the lateral aspects of the anterior pituitary gland that synthesize and release growth hormone (somatotropin). These cells represent the most abundant cell type in the anterior pituitary, accounting for approximately 50% of all pituitary cells. Somatotroph function is regulated by growth hormone-releasing hormone (GHRH) and somatostatin from the hypothalamus. Dysfunction of somatotrophs can lead to growth hormone deficiency resulting in short stature, or excess growth hormone production causing gigantism in children. Understanding somatotroph physiology is essential for managing growth disorders in pediatric patients.
