An oncometabolite is a metabolic intermediate that accumulates to abnormally high levels in cancer cells due to mutations in metabolic enzymes, subsequently contributing to tumor development and progression. The most well-characterized oncometabolites include 2-hydroxyglutarate (produced by mutant IDH1/2 enzymes), succinate (from succinate dehydrogenase mutations), and fumarate (from fumarate hydratase mutations). These compounds can act as competitive inhibitors of α-ketoglutarate-dependent dioxygenases, leading to widespread epigenetic changes including DNA and histone hypermethylation. Oncometabolites represent a direct link between metabolic dysfunction and cancer biology, offering both diagnostic biomarkers and potential therapeutic targets. Detection of oncometabolites can guide treatment decisions, particularly in brain tumors and certain sarcomas.
