A specialized laboratory test that evaluates sperm chromatin structure and DNA integrity using flow cytometry and acid denaturation. The assay measures the susceptibility of sperm DNA to acid-induced denaturation, providing information about DNA fragmentation index (DFI) and high DNA stainability (HDS). Elevated DFI values are associated with reduced fertilization rates, poor embryo quality, increased miscarriage rates, and failed assisted reproduction cycles. The test is particularly useful in cases of unexplained infertility, recurrent pregnancy loss, or repeated ART failures. Normal DFI values are typically below 15%, with values above 30% indicating significant DNA damage.
