oncogenic fusion protein

A chimeric protein resulting from chromosomal rearrangements that combines functional domains from different genes to drive malignant transformation.

Full Definition

An oncogenic fusion protein is created when chromosomal translocations, inversions, or other rearrangements join portions of two different genes, resulting in a novel protein with altered function that promotes cancer development. These fusions typically combine regulatory domains from one protein with catalytic or DNA-binding domains from another, creating constitutively active or mis-regulated proteins. Common examples include BCR-ABL in chronic myeloid leukemia and EWS-FLI1 in Ewing sarcoma. In cancer genomics, detection of specific fusion proteins is crucial for diagnosis, prognosis, and targeted therapy selection. RNA sequencing and specialized fusion detection algorithms are typically required to identify these aberrant transcripts.
