Full Definition

Mantle cell lymphoma is a distinct B-cell neoplasm characterized by the t(11;14)(q13;q32) translocation leading to cyclin D1 overexpression. The lymphoma cells originate from the mantle zone of lymphoid follicles and typically show a diffuse growth pattern, though nodular and mantle zone patterns may be seen. The disease has an aggressive clinical course with a tendency for widespread dissemination at presentation, frequently involving extranodal sites including the gastrointestinal tract and bone marrow. Treatment typically involves intensive immunochemotherapy regimens, and autologous stem cell transplantation may be considered in appropriate candidates.
