Full Definition

The TREC assay (T-cell Receptor Excision Circles assay) is a quantitative PCR-based test used in newborn screening to detect severe combined immunodeficiency (SCID) and other T-cell lymphopenias. TRECs are stable DNA byproducts formed during T-cell development in the thymus. Low or absent TREC levels indicate impaired T-cell production, suggesting primary immunodeficiency disorders. The assay is performed on the same dried blood spot used for other newborn screening tests, making it an efficient addition to screening panels. This test has revolutionized early detection of SCID, enabling life-saving interventions before the onset of serious infections.
