Melanocortin receptors (MCRs) are a family of five G-protein coupled receptors (MC1R-MC5R) that bind melanocortin peptides derived from pro-opiomelanocortin (POMC), including α-MSH, β-MSH, γ-MSH, and ACTH. Each receptor subtype has distinct tissue distribution and physiological functions: MC1R regulates skin pigmentation, MC2R mediates ACTH effects on adrenal steroidogenesis, MC3R and MC4R are crucial for energy homeostasis and feeding behavior in the hypothalamus, and MC5R has roles in exocrine gland function. Dysfunction of melanocortin receptors, particularly MC4R, is associated with severe obesity, while MC2R mutations cause familial glucocorticoid deficiency. These receptors are important therapeutic targets for metabolic disorders.
